Chemical constituents and anticancer activity from the soft coral of
Lobophytum crassum

Abstract
This study investigates the potential anti-cancer properties of secondary metabolites from the
ethyl acetate extract of the soft coral Lobophytum crassum from Taiwanese waters. The coral
species was identified using morphology, sclerites, and DNA sequencing (mtMuts, COI, and
28S rDNA). Separation using medium-pressure liquid chromatography followed by RP-HPLC
purification of the extract was carried out, resulting in the isolation of six compounds 1–6. Their
structures were elucidated by spectroscopic analysis including NMR, MS, IR, and optical
rotational data to be one polyhydroxysteroid, three cembranoids, one monoterpenoid lactone,
and one carotenoid, and were named 3β,22,23β -cholesta-5,17(20)-dien-3,22,23,25-tetrol (1),
isosarcophine (2), trocheliolide B (3), crassumol A (4), loliolide (5), and peridinin (6). Of these,
compound 1 was previously reported from Sinularia mayi, but this is the first isolation from L.
crassum. Compound 2 showed moderate anticancer activity in MDA-MD-231.
