Effect of Scutellaria barbata D. Don on bladder cancer cells and macrophages

Abstract
In this study, we identify the effect of a herbal medicine Scutellaria barbata D. Don on the
interaction of bladder cancer cells and their surrounding macrophages. Scutellaria barbata D.
Don belongs to the category of heat-cleaning drugs in traditional herbal medicine. It is known
that there are nearly 300 compounds in Scutellaria barbata D. Don, primarily flavonoids and
diterpenes. For functional studies, it has been found to have immunomodulatory, anti-tumor,
antibacterial and anti-viral activities. In our study, we found that the water extracts of
Scutellaria barbata D. Don have cytotoxicity in bladder cancer cells 5637 and BFTC909 in a
dose-dependent manner. Interestingly, there is no cytotoxicity to macrophages at the same dose
(4 mg/ml). In the cancer cell migration assay, 4 mg/ml of Scutellaria barbata D. Don water
extracts couldn’t inhibit the migration rate of 5637 cells after 6-h treatment. But the conditioned
medium of M1 macrophages inhibited the migration rate of 5637 cells. Furthermore, we found
that Scutellaria barbata D. Don water extracts could inhibit the expression of M2 macrophage
markers (MRC-1 and CCL17) and some M1 macrophage markers (IL6 and IL-12β), but not the
M1 macrophage markers (CCL2 and TNFα). It indicates that Scutellaria barbata D. Don water
extracts have the ability to decrease the differentiation of M2 macrophages. Based on these
results, it suggests that Scutellaria barbata D. Don water extracts inhibit bladder cancer cell
proliferation. M1 macrophage conditioned medium inhibited 5637 cell migration, and
Scutellaria barbata D. Don tended to reduce M2 macrophage differentiation.
